Abstract
This study was performed to investigate direct effects of macrophage-derived cytokines, transforming growth factor-beta 2 [TGF-beta 2] and tumor necrosis factor-alpha [TNF-alpha],on the chemotactic migration of choroidal endothelial cells [CECs]. Bovine CECs were grown to subconfluency [passage 2 to 4] and pretreated with TGF-beta 2 [1, 5, and 10ng/ml] or TNF-alpha [1, 5, and 10ng/ml]. Migration was determined by a modified Boyden chamber assay using 5 x10^5 bovine CECs in the upper chamber, 8micrometer pore inserts coated with fibronectin, and VEGF [10ng/ml] added in the lower chamber as a chemotactic factor. TGF-beta 2inhibited migration of CECs by 14.5% at high concentration [10ng/ml], but had no effect at the other concentrations. TNF-alphainhibited migration of CECs by 28.6% at high concentration [10ng/ml], but stimulated migration by 17% at low concentration [1ng/ml]. Our results showed that macrophage derived cytokines, TGF-alphaand TNF-alphamight have complex effects on the chemotactic migration of bovine CECs induced by VEGF.
